The deformation on cylindrical surfaces of holes in tensile loaded laminated composite specimens was measured using new moire interferometry techniques. These new techniques were developed and evaluated using a 7075-T6 aluminum control specimen. Grating replication techniques were developed for replicating high quality diffraction gratings onto the cylindrical surfaces of holes. Replicas of the cylindrical specimen gratings (undeformed and deformed) were fabricated onto circular steel sectors. Narrow angular regions of these sector gratings were directly evaluated in a moire interferometer. This moire interferometry approach eliminated potential sources of error associated with other moire interferometry approaches.

Juvenile hormone (JH) plays pivotal roles in the development and reproduction of insects. Efforts to characterize the mechanisms of JH regulation are complicated due to JH pathways often being intertwined with those of 20-hydroxyecdysone (20E). Upon adult emergence, female Aedes aegypti enter a period of development during which they gain competence for mating, bloodfeeding, and egg production. JH levels rise dramatically and peak during the first 2-3 days post-emergence and remain relatively high until a bloodmeal is consumed, while 20E titers remain very low throughout the entire stage. Thus, post-emergence development offers a unique opportunity to study the effects of JH in the absence of 20E. The purpose of this study was to begin to understand what exactly occurs when young children and adult day care participants interact together during planned intergenerational activities. Data were recorded through field notes, video tapes, activity plans and evaluations and through the assessment of the interaction by using an adaptation of the Early Childhood Classroom Observation Scale. The presentation of discoveries includes a description of each of the 12 activities implemented between the two groups, the environment, interactions between staff and participants and interactions between children and adults. The findings indicate implications for practice as well as future directions for research.

In this thesis, we focus on the steady state properties of two systems which are genuinely out of equilibrium. The first project is an application of dynamic field theory to a specific non equilibrium critical phenomenon, while the second project involves both simulations and analytical calculations. The methods of field theory are used on both these projects. In the first part of this thesis, we investigate a generalization of the well-known field theory for directed percolation (DP). The DP theory is known to describe an evolving population, near extinction. We have coupled this evolving population to an environment with its own nontrivial spatio-temporal dynamics. Open reading frame (ORF) sso0433 from the archaeon Sulfolobus solfataricus encodes a protein kinase, SsoPK5 that exhibits 33% sequence ident ity to p53 related protein kinase (PRPK) from Homo sapiens and 26% sequence identity to piD261/Bud32 from Saccharomyces cerevisiae. Given this high degree of similarity, the objectives of this thesis were to (a) clone and purify recombinant SsoPK5, (b) examine its commonalities and differences with its eukaryotic homologues, and (c) determine if it was regulated by nucleotides or related compounds. Substantial progress was achieved on each objective. We investigate the nonequilibrium steady state of driven magnetic flux lines in type-II superconductors subject to strong point or columnar pinning centers and the aging dynamics of nonequilibrium relaxation process in the presence of weak point pinning centers. We employ a three-dimensional elastic line model and Metropolis Monte Carlo simulations. For the first part, we characterize the system by means of the force-velocity / current-voltage curve, static structure factor, mean vortex radius of gyration, number of double-kink and half-loop excitations, and velocity / voltage noise features. We compare the results for the above quantities for randomly distributed point and columnar defects. The glpX gene of Escherichia coli encodes fructose 1,6-bisphosphatase II (FBPase II), an enzyme that would appear to be redundant with FBPase I, encoded by fbp. However, glpX mutants have no apparent phenotype, while fbp mutants are unable to grow on gluconeogenic substrates as sole carbon sources, suggesting that GlpX function is insufficient for growth of fbp mutants under these conditions. To gain insight into the physiological functions of the FBPases, regulation of glpX expression was investigated. It was found that glpX is transcribed as part of a complex glpFKX operon containing promoters upstream of glpF, glpK and glpX (PglpF, PglpK, PglpX, respectively). The effect of Mn(III)-salophen, a superoxide scavenger, against oxidative stress was evaluated in Escherichia coli sodA- sodB-. Oxidative stress was imposed by exposure of the cells to paraquat or hyperoxia. Cells were grown in LB medium overnight, washed and resuspended in the indicated glucose/salts medium supplemented with casamino acids. The effect of Mn(III)-salophen in the oxidative stress model in vivo was measured in terms of the cell growth. Mn(III)-salophen ( 60 nM) completely protected E. coliJI132sodA- sodB-against 1.0 μM paraquat. Equivalent amounts of Mn(III) acetate, a Mn(III)-salophen component, also protected against paraquat toxicity in aerobic E. coli JI132sodA- sodB-.
